چکیده

The aim of this study was to assess how cerivastatin influences contractility and degree of myocardial damage in papillary muscle subjected to hypoxia-reoxygenation in hypercholesterolemic guinea pigs. Study group consisted of guinea pigs, fed standard, hypercholesterolemic or hypercholesterolemic diets with low dose of cerivastatin. During experimental hypoxia-reoxygenation, the contractility was measured. Apoptosis of cardiomioctes was assessed with the use of TUNEL technique. Total cholesterol in standard, hypercholesterolemic and cerivastatin-treated group was 35 +/- 8 mg/dl, 131.6 +/- 30.4 mg/dl and 121.2 +/- 26.2 mg/dl, respectively, and was significantly higher in rats fed hypercholesterolemic and hypercholesterolemic + cerivastatin diets than in control group (p < 0.01). There were no significant differences between all analyzed groups in the post-ischemic cardiac function. Percentage of apoptotic cells after hypoxia-reoxygenation injury in groups fed standard, hypercholesterolemic and hypercholesterolemic + cerivastatin diets was 30 +/- 8%, 20 +/- 4% and 5 +/- 7%, respectively, and was significantly lower in groups that received hypercholesterolmic (p < 0.01) and hypercholesterolemic + cerivastatin (p < 0.001) diets in comparison with standard diet-fed group. In the group treated with cerivastatin, the percentage of apoptotic cells was additionally lower in comparison with hypercholesterolemic group (p < 0.01). Negative correlation between percentage of apoptotic cells and HDL level was found when all groups were considered jointly (r = -0.41, p < 0.05). Our study clearly shows that cerivastatin in hypercholesterolemic animals and hypercholesterolemia itself limit cardiomiocyte damage after hypoxia-reoxygenation.

Protection by hypoxic preconditioning against hypoxia-reoxygenation injury in guinea-pig papillary muscles.

OBJECTIVE Developed tension in guinea-pig papillary muscles is depressed by prolonged hypoxia; subsequent reoxygenation leads to a partial recovery that stabilizes after an early period of arrhythmia. We have investigated whether hypoxic preconditioning in these muscles (1) improves the recovery of developed tension, (2) protects against arrhythmia, and (3) causes other significant electromecha...

Electrical heterogeneity and conduction block in reoxygenated guinea pig papillary muscles.

To investigate the changes in electrical activity after reoxygenation, guinea pig papillary muscles were reoxygenated with 5 mM glucose solution after various durations (30-120 min) of substrate-free hypoxia. Action potential duration recovered to the control level on reoxygenation following 30-120 min hypoxia. The recovery of developed tension or resting tension was incomplete after reoxygenat...

Arrhythmic activity in reoxygenated guinea pig papillary muscles and ventricular cells.

Aftercontractions, delayed afterdepolarizations, and automaticity occurred in guinea pig papillary muscles that were reoxygenated after hypoxic conditioning. The emergence of dysfunction was dependent on the severity of hypoxic conditioning and on stimulation during reoxygenation. After 60 minutes of substrate-free hypoxia, reoxygenation induced automaticity in a high proportion of stimulated m...

Insulin Protects Apoptotic Cardiomyocytes from Hypoxia/Reoxygenation Injury through the Sphingosine Kinase/Sphingosine 1-Phosphate Axis

OBJECTIVE Experimental and clinical studies have shown that administration of insulin during reperfusion is cardioprotective, but the mechanisms underlying this effect are still unknown. In this study, the ability of insulin to protect apoptotic cardiomyocytes from hypoxia/reoxygenation injury using the sphingosine kinase/sphingosine 1-phosphate axis was investigated. METHODS AND RESULTS Rat ...

Silver nanoparticle induced muscle abnormalities : A sub-chronic dermal assessment in guinea pig

Nanosilver has recently been recognized as an antimicrobial agent. Although this nanoparticlecan be used in medical applications but its dermal and systemic toxicity via dermal exposure hasnot been completely determined yet. The aim in this study was to investigate the potential dermaltoxicity of Nanosilver in subchronic method. Before the colloidal silver nanoparticle toxicityevaluation, their...
